Nos projets de recherche concernent l’architecture moléculaire et les mécanismes fonctionnels d’enzymes agissant sur les glycanes (CAZymes), ainsi que d’enzymes, récepteurs/canaux et molécules d’adhésion cellulaire d’intérêt neurobiologique.

Les CAZymes gouvernent l’hydrolyse, la conversion ou la reconnaissance de glycanes issus de glycoconjugués d’origines très variées comme les animaux, les champignons, les plantes et bactéries. Ces composés sont impliqués dans la majorité des processus biologiques fondamentaux tels que le développement, le contrôle qualité lors du repliement des protéines, la reconnaissance hôte-pathogène, la réponse immunitaire, et peuvent être associés à des maladies telles que les maladies métaboliques ou neurodégénératives. En collaboration avec l’équipe “Glycogénomique”, nous nous focalisons sur plusieurs familles de glycosides hydrolases dont certaines ont un intérêt pour des applications biomédicales ou biotechnologiques. Nous étudions aussi certaines enzymes de maturation des glycosaminoglycanes tels que l’héparane sulfate impliqué dans de nombreux processus physiologiques et pathologiques.

Nous nous intéressons également à la fonction, aux propriétés de reconnaissance et à la structure de récepteurs et molécules d’adhésion impliqués dans la formation et le fonctionnement des systèmes nerveux central et périphériques, avec un intérêt particulier pour leurs relations structure-fonction, leur spécificité de reconnaissance des partenaires et ligands, leur dynamique conformationnelle et leur régulation fonctionnelle. Dans ce contexte, nos molécules modèles sont l’enzyme acétylcholinestérase et le domaine extracellulaire du récepteur nicotinique à l’acétylcholine, deux protéines pour lesquelles certaines anomalies d’origine génétique ou dysfonctionnements sont corrélés à des maladies neuro-dégénératives, ainsi que les neuroligines, des molécules d’adhésion cellulaire structuralement (mais pas fonctionnellement) apparentées à l’acétylcholinesterase, et pour lesquelles certaines mutations sont associées à des maladies neuro-développementales..

Nos recherches portent également sur l’étude structurale et fonctionnelle de protéines senseurs membranaires impliquées dans la formation de biofilm chez le pathogène Pseudomonas aeruginosa , ou encore sur des enzymes impliquées dans le métabolisme lipidique chez le pathogène Mycobacterium tuberculosis.

Notre approche méthodologique requiert l’expression quantitative de protéines recombinantes solubles dans des systèmes procaryotes ou eucaryotes et l’utilisation d’approches complémentaires en biochimie et biophysique pour leur caractérisation fonctionnelle et structurale.
