Notre équipe se focalise sur la compréhension des mécanismes d’infection virale et pseudo-virale, principalement régis par l’interaction de protéines virales, de facteurs cellulaires et d’acides nucléiques assemblés en complexes multifonctionnels. Dans certains cas, comme pour de nombreux virus à ARN à brin positif, la réplication est associée à la génération d’organelles membranaires où se produisent la réplication et la transcription. Notre objectif est de comprendre les mécanismes par lesquels les multiples activités enzymatiques sont coordonnées entre elles et régulées pour réaliser efficacement l’infection. À cette fin, la compréhension des interactions des protéines virales avec les facteurs protéiques cellulaires et les membranes est cruciale. Pour atteindre nos objectifs, nous utilisons un large éventail de techniques biochimiques et biophysiques combinées à la cristallographie aux rayons X et à la microscopie électronique.

Complexes de réplication des alphavirus :

L’objectif principal de nos recherches est la caractérisation fonctionnelle et structurale du complexe de réplication des alphavirus. Les alphavirus, comme le virus du chikungunya (CHIK) ou le virus de l’encéphalite équine de l’Est (EEEV), provoquent des maladies infectieuses zoonotiques à fort impact en santé humaine. La réplication du génome viral et l’expression des gènes sont réalisées par complexe de réplication virale multifonctionnel associé aux membranes.

Notre but est de caractériser au niveau fonctionnel et structural les différents états de ce complexe de réplication pour proposer un modèle mécanistique complet décrivant à l’échelle moléculaire le cycle d’infection viral et ses mécanismes de régulation. Les résultats devraient dévoiler des informations précieuses et permettre de proposer de nouveaux outils pour contribuer au développement de nouvelles stratégies thérapeutiques pour le traitement de ces maladies infectieuses.

Mécanisme de réplication des bunyavirus :

Les Bunyavirales constituent un ordre très vaste et diversifié de virus à brin négatif segmenté (sNSV) comprenant plus de 350 espèces classées dans de nombreuses familles dont les Hantaviridae et les Nairoviridae, qui comptent plusieurs virus hautement pathogènes pour l’homme. Il est donc nécessaire de (i) comprendre leur mécanisme d’infection et (ii) de développer des médicaments efficaces pour leur treaitement.

Dans ce contexte, nous étudions deux étapes critiques du cycle viral des bunyavirus : la réplication et la transcription. Ces réactions sont réalisées par la polymérase virale multifonctionnelle (L) qui interagit spécifiquement avec les extrémités 3′ et 5′ des segments d’ARN viral. Alors que l’initiation de la réplication est réalisée de novo, sans nécessiter d’amorce, l’initiation de la transcription est réalisée par le mécanisme unique de “cap-snatching”. Les structures à résolution atomique de l’Infuenza A et du LACV nous ont permis de proposer le premier modèle complet basé sur la structure de la réplication des bunyavirus, qui pourrait être applicable à d’autres NSV, mais qui a également soulevé de nouvelles questions essentielles. Par conséquent, nous visons la caractérisation structurelle et fonctionnelle des polymérases des bunyavirus.

Intégration des rétrotransposons de la levure :

Les rétroéléments se répliquent par transcription inverse de leur génome ARN en un ADNc qui est intégré de manière stable dans le génome de la cellule hôte par leur propre intégrase (IN). L’intégration ne se produit pas de manière aléatoire in vivo, révélant un modèle de sites préférés spécifique au rétroélément, qui dépend principalement de l’interaction de l’IN avec des facteurs cellulaires qui fixent l’intégration à des sites spécifiques. Cependant, les mécanismes par lesquels ces facteurs interagissent avec les IN et contribuent au processus d’intégration sont encore mal compris. Des connaissances importantes sur la biologie des rétrovirus ont été acquises en étudiant les rétrotransposons Ty LTR de la levure. Ty1 cible son intégration sur les gènes transcrits par l’ARN polymérase III (Pol III), qui sont des régions pauvres en gènes du génome de la levure. Cette remarquable sélectivité du site d’intégration a été bien étudiée in vivo et in vitro. Cependant, il n’existe aucune information structurelle sur le processus d’intégration ou l’interaction avec Pol III. Dans ce projet, nous chercherons à savoir quels sont les mécanismes d’intégration et comment la protéine Pol III s’intègre.
